Abstract

Rabbits’ eyes were exposed to argon fluoride excimer laser 193 nm using Lambda Physik laser instrument, the pulse rate was 0-200 Hz, the optical zone was 1cm2, the energy/pulse was 2.0 mJ and the pulse duration was 6 ns. Two different fluencies (300 mJ/cm2 and 500 mJ/cm2 were applied to design the side effect of this wavelength on the aqueous humor and lens of eye rabbits. Twenty New Zealand rabbit were used in this study where they were divided into four groups.In the 1st group the right eyes were submitted to 300 mJ/cm2 and the animals were left for 1 week, the 2nd group was submitted to 300 mJ/cm2 and was left for 4 weeks, the 3rd group was submitted to 500 mJ/cm2 and was left for 1 week and finally the 4th group was submitted to 500 mJ/cm2 and was left for 4 weeks. The left eyes from each group were used as control.Measurements of the total protein concentration, the refractive index (RI), column chromatography, sodium dodecyle sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and ultraviolet spectroscopy were investigated after 1 and 4 weeks of treatment with 193 nm ArF excimer laser.In the aqueous humor samples, non-significant changes in the total protein concentration and the refractive index (RI) were observed after 1 week of treatment with 300 mJ/cm2. All measurements detected significant changes after 4 weeks of treatment with 300 mJ/cm2 and after 1 and 4 weeks of laser treatment with 500 mJ/cm2. The ultraviolet absorption spectrum revealed apparent depletion in ascorbic acid concentration.The result of lenses samples indicates significant alteration in lens protein concentration and the refractive index (RI) measurements. The electrophoresis mobility and column chromatography separation indicates significant changes in molecular weights of lens protein. The ultraviolet absorption spectrum revealed apparent depletion in lens protein concentration with pronounced changes outside the range of protein absorbance.It was concluded that, ArF excimer laser (193 nm) leads to change in aqueous humor and formation of new protein with different molecular weights accompanied with a loss and oxidation of ascorbic acid and formation of dehyroascorbic acid and 193 nm ArF excimer laser treatment has a direct deleterious effect on lens protein which may be a possible risk for cataractogenes.
