Abstract

The present study was carried out to evaluate the effect of chamomile on tongue wound healing in alloxan-induced diabetic rats using light and transmission electron microscopes. In addition histomorphometric study using image J analysis was used. Forty-five rats were used in this study and they were divided into three groups, each group consists of 15 rats. They randomly classified into 3 equal groups: control group, diabetic untreated group and diabetic treated with chamomile group. Diabetes was induced by single intraperitoneal injection of 50 mg/kg Monohydrated Alloxan dissolved in sterile 0.9% saline (10 mg/ml). Oral mucosal wound was done on the dorsal surface of tongue of all rats in the central region, 3 mm from the tip with 1 mm in depth. 5 rats from each group were euthanized at 7th, 14th and 28th days after wounding. The tongue of each rat was dissected carefully and prepared for qualitative histological studies using Light & Transmission Electron microscopes and for quantitative assessment. Light microscopic examination: The samples were fixed for 2 days at room temperature in 10% formalin. Then were dehydrated in graded alcohols, embedded in paraffin, and cut serially into 5 μm in middle sagittal sections. The sections will be stained with:*Hematoxylin and Eosin (H&E) and *Masson’s Trichrome (MTC). Electron microscopic examination: The other samples were trimmed into cubic 1 mm and fixed in mixture of 4% paraformaldhyde and 1%glutaraldhyde for 24 hours. Then, they were processed for TEM examination.
