Abstract
Identification is necessary in mass disaster. This identification can be done by different methods but the most important method is DNA analysis. For successful identification, we need to preserve this DNA from degradation. For this purpose, samples should be preserved in refrigerators at certain cool temperatures and this not available in most times due to shortage of resources. So in the present study we search for simple, efficient, available and not expensive methods of preservation of human remains to be used in identification especially most mass disasters occur in remote areas from laboratories. We used for this purpose three preservatives (DMSO, NaCl, Ethanol70%).
This study aimed to provide an effective method for preservation of human tissues on quality and quantity of DNA to be used in DVI and this can be achieved by:
- Comparing between the effect of different preservative agents [DMSO, NaCl, Ethanol70%] on quality and quantity of DNA extracted from human muscle tissues.
- Comparing the effect of time on quality and quantity of DNA extracted from human muscle tissues with using different preservative agents [DMSO, NaCl, Ethanol70%].
This study was conducted at the clinical pathology department in faculty of medicine-Suez Canal University (FOM-SCU).
• Muscle samples were collected from patients admitted to Suez Canal University Hospital for surgical amputation of part of limb (normal healthy muscle tissues) after taking their consent and consent approval of Research Ethics Committee (REC) of FOM-SCU).
Collected tissues were placed in clean plastic bags and were set on ice, then returned to the laboratory and placed at -20 ºC and processed the following day.
