Abstract

The present study aiming to determine the biodiversity of Rhizobium
leguminosarum symbiovar Viciae at some Egyptian governorates which its soil
varied at fertility, salinity and pesticides applications. To achieve this goal;
thirteen Rhizobia isolates were collected from ten governorates and subjected
to the following biological assays:
 Growth rate and morphological character levels, i.e. salinity tolerance,
herbicide resistance, antibiotics resistance and carbon source
utilization.
 Biochemical differentiation levels such as indol acetic acid production,
catalase enzyme and protein banding pattern.
 Molecular level such as plasmid profile, Nod c gene detection and
RAPD – PCR for its DNA content.
The principle objectives of this study are isolation salt and herbicide tolerant
Rhizobium which can help their host plant to support adverse ecological
conditions and investigate the effects of antibiotics, herbicide and salt on
growth of Rhizobium isolates. Furthermore detect the diversity of these
populations by using several biochemical and molecular approaches to observe
their diversity and their relationship with different isolates.
