Abstract

Objectives: The present study aimed to assess the antifungal effect of silver nanoparticles(SNPs) against induced oral candidiasis in albino rats,as well as to compare the antifungal activity of silver nanoparticle solution and miconazole oral gel.
Materials and Methods: One hundred and sixty male albino rats were included. After induction of oral candidiasis using Candida albicans (ATCC 90028), rats were randomly divided into the following experimental groups; 32 rats each. Group1 (normal (–ve) control group): rats without oral candidiasis (not immunosuppressed , neither infected or drug treated). group 2 (+ve control group): rats with oral candidiasis (immunosuppressed and infected but not treated). group 3 (silver nanoparticles 50 μg/ml group): rats treated topically with silver nanoparticle solution (50 μg/ml/day/2 weeks). group 4 (silver nanoparticles100μg/ml group): rats treated topically with SNPs solution (100 μg/ml/day/2 weeks). group 5 (miconazole gel group): rats treated topically with miconazole 2% oral gel (10 mg/kg/day/2 weeks). Baseline assessment was conducted on day 0 (just before drug treatment). Evaluation of the candidal lesions during drug treatment was conducted on day 7, day 14 and the follow up assessment on day 30. At all-time points, clinical evaluation was performed to evaluate the extent of oral candidal lesions. Direct examination of fungal cultures was performed at all time points for microbiological study. Eight rats from each group were sacrificed at all time points for tongue biopsies and compare the histological changes between different groups.For toxicity evalution,blood samples were taken for measuring liver and kidney enzymes.Also, liver and kidney tissue organs of all rats were studied.
Results: Silver nanoparticle solution had superior antifungal effect compared to miconazole oral gel in terms of lesion size reduction, microbial counting as well as histopathological changes.
Conclusion: Silver nanoparticle solutions in a concentration of 50 and100 μg/ml are promising candidates for treatment of oral candidiasis without toxicity to liver or kidney tissues or enzymes.
