Abstract

Ferric nitrilotriacetate (Fe-NTA) is a known potent nephrotoxic agent. The present investigation, clarify the chemo-preventive and ameliorative effects of Hibiscus rosa sinensis extract (HRE) on Fe-NTA-induced renal iron overload toxicity and its consequent oxidative stress. 30 male albino rats were divided into 5 groups, 6 rats each. Group I, served as control group, animals received only saline solution injection i.p., at a single dose of 10 mg/kg b.w. Group II, animals were given HRE 200 mg/kg, b.w., orally, for 7 days). Group III,animals received a single i.p. dose of Fe-NTA 9 mg Fe/kg b.w.). Group IV, animals were pre-treated with HRE, as group II, followed by single i.p. dose of Fe-NTA, as group III.Animals of group V received a single dose of Fe-NTA followed by post-treatment with HRE,at the same doses. The results indicated a marked deterioration of renal function as evidenced by a sharp increase in blood urea and serum creatinine was observed (183.9%
and 195.4%, respectively) after injection of Fe-NTA. On the other hand, Fe-NTA induced a significant renal oxidative stress demonstrated by reduction in activities of renal catalase, superoxide dismutase and Glutathione-s-transferase (61.5%, 42.8%, and 56.3%, respectively). While renal xanthine oxidase activity and serum advanced oxidation protein products elevated significantly (128.8% and 89.3%, respectively). Both mode of administration,pre- and post-treatment with HRE resulted in a signi􀂿cant protection and amelioration for blood urea, serum creatinine, xanthine oxidase (P < 0.001), and advanced oxidation protein products (P < 0.01). HRE was also capable for restoring the renal antioxidant enzymes catalase, superoxide dismutase and Glutathione-s-transferase, nearly, to normal< level. It was noted that animals treated with HRE prior to Fe-NTA was more effective.The present data suggest that Hibiscus rosa sinensis may be valuable as an effective preventive and/or ameliorative agent against Fe-NTA-mediated iron overload and its renal oxidative stress and toxicity.
