Abstract Two field experiments were conducted during the seasons of ٢٠٠١/٢٠٠٢ and ٢٠٠٢/٢٠٠٣ at the farm of Minufiya University in El Sadat City (under furrow irrigation system). The aim of this study was to investigate the effect of biofertilizers (Non – inoculation, Rizobactrein, Cerealin and Nitrobin), nitrogen levels ( ٠ kg/fad, ٤٠kg/fad, ٦٠ kg/fad and ٨٠ kg/fad )and their interactions on yield, and quality characters of five sugar beet (Beta vulgaris, L.) cultivars. We also used RAPD for genetic variation and phylogenetic relationships among genotypes belonging to five sugar beet (Beta vulgaris, L.) cultivars (DEL ٩٣٦ ET, A – ٠٠٣٤, FD ٩٩٠١, Kaweterma and Lola) which used in present study. The results obtained could be summarised as follows: Inceasing the levels of nitrogen fertilizer increased the values of root weight in kg/plant, top weight in g/plant, root yield in ton/faddan and top yield in ton/faddan. Inoculation with Cerealin increased significantly top weight in g/plant and top yield in ton/faddan. Sugar yield in ton/faddan, root weight in kg/plant and root yield in ton/faddan were increased significantly under inoculation with rizobactrein. Cultivar Kaweterma gave the highest mean value of root weight in kg/plant, root yield in ton/faddan and sugar yield in ton/faddan. And cultivar DEL ٩٣٦ gave the highest mean value of top weight in g/plant and top yield in ton/faddan. Inceasing the levels of nitrogen fertilizer decreased the values of sucrose percentage, juice purity percentage, amino-N and incresed percentages of potassium (K %) and sodium (Na %). Inoculation with Cerealin increased significantly amino-N and percentage of potassium (K %) and sodium (Na %) / amino-N. Total soluble solids (TSS %), sucrose percentage, juice purity percentage, and were increased significantly under inoculation with Rizobactrein. Cultivar Kaweterma gave the highest mean value of juice purity percentage and amino-N. And cultivar FD ٩٩٠١ gave the highest mean value of Total soluble solids (TSS %) and sucrose percentage. The results revealed that, the five primers scored a total of ١١٥ amplified DNA bands. The number of bands varied from ٢٠ bands (OPZ-١٢) and (OPZ-٢٠) to ٢٩ bands (OPC-١٠). It may be cocluded that primer OPC-١٠ scored the largest number of RAPD markers against agarose matrix. While the primers OPZ-٢٠ and OPZ-١٢ recorded the lower number of RAPD markers against agarose matrix. The number of amplified scored fragments per cultivars ranged from ١٧ to ٢٤ bands. A-٠٠٣٤ cultivar scored the largest number of amplified fragments. While the FD ٩٩٠١ cultivar recorded the lower numbe of amplified fragments. The OPC-١٠ have the highest reaction facilities in all accession of all five sugar beet cultivars to produce the highest number of amplified RAPD-PCR bands. A genetic similarity among four cultivars ranged from ٣٨.٦% between Kaweterma and Lola to ٥٩.١% between DEL٩٣٦ET and A-٠٠٣٤. Dendrogram constructed with UPGMA cluster analysis of RAPD data showing the genetic relationships among five sugar beet cultivars and illustrated that RAPD data distinguished two main clusters. The first cluster included the accessions of DEL ٩٣٦ ET, A-٠٠٣٤ and Lola. While the second main cluster included the accessions of FD ٩٩٠١ and Kaweterma. The first main cluster divided into two sub-cluster, the first one had the accessions of DEL ٩٣٦ Et and A-٠٠٣٤. The other sub-cluster included the accessions of Lola. The accessions of DEL ٩٣٦ ET, A-٠٠٣٤ and Lola were more distances, while the accession of FD ٩٩٠١ was more related to the accession Kaweterma. Generally, we recommended by cultivars Kaweterma and FD ٩٩٠١ under application of ٦٠ kg/fad. mineral nitrogen with Cerealin or Rizobactrein as biofertilizater to obtain high sugar beet yield, yield components and quality. |
