Abstract

Spices and herbal remedies have been used since ancient times to treat a variety of disorders. It has been experimentally demonstrated
that spices, herbs, and their extracts possess antimicrobial, anti-inflammatory, antirheumatic, lipid-lowering, hepatoprotective, nephroprotective, antimutagenic and anti-ulcer activities. So this study aimed to identify the chemical composition of Curcuma longa (Turmeric) and Nigella sativa (black seed) and fatty acid analysis of black seed oil. Study the effect of different treatment of turmeric and
black seed (2.5% turmeric, 5% turmeric, 2.5 black seed, 5% black
seed, 2.5% turmeric & black seed, 5% turmeric & black seed) on
gastric ulcer including pH value, volume of gastric juice, total acidity,
ulcer index, body weight, feed intake, feed efficiency ratio, some
organs weight and histopathology examination of stomach.
Male albino rats (56 rats), Sprague Dawely strain, Weighting
(120 – 125 g) were used. All rates (56 rats) were fed on standard diet
for one week before starting the experiment for acclimatization. This
rats were divided into 9 groups each group 7 rats. The first two
groups (1, 2) were fed on standard diet for another three weeks.
While the remaining groups (3, 4, 5, 6, 7, 8) were fed on the
experimental diet for three weeks.
After the three weeks, all rats except groups (1) were fasted for
24 hours and allowed to drink water only. In the morning of the next day all rats were given aspirin orally (200 mg / kg b.wt) to induce
gastric ulcer. After that in the morning of the next day each group
was given its diet until the end of the fourth week.
 Group (1): Rats fed on standard diet only as a control negative
(healthy group).
 Group (2): Rats fed on standard diet only as a control positive
(given aspirin).
 Group (3): Rats fed on standard diet containing 2.5% of turmeric
(powder) replaced by corn starch.
 Group (4): Rats fed on standard diet containing 5% of turmeric(powder) replaced by corn starch. Group (5): Rats fed on standard diet containing 2.5% of black
seed (milled)replaced by corn starch.
 Group (6): Rats fed on standard diet containing 5% of black seed
(milled) replaced by corn starch.
 Group (7): Rats fed on standard diet containing 2.5% turmeric
(powder) and black seed (milled) (1 : 1) replaced by corn starch.
 Group (8): Rats fed on standard diet containing 5% turmeric
(powder) and black seed (milled) (1 : 1) replaced by corn starch The obtained results can be summarized as follows:
1. Chemical composition of turmeric:
Results showed that, chemical composition of turmeric were
moisture (8.96%), fat (5.32%), protein (6.14%), ash (4.43%), crude
fiber (7.24%), total carbohydrate (67.91%).
2. Chemical composition of black seed:
Chemical composition of black seed were moisture (6.13%), fat
(33.57%), protein (21.08%), ash (4.43%), crude fiber (5.48%), total
carbohydrate (29.31).
3. Fatty acid analysis of black seed oil:
Fatty acid analysis of black seed oil were myristic acid C14:0
(1.28%), palmitic aicd C16:0 (14.26%), oleic acid C18:0 (22.05%), stearic
acid (3.17%).
4. Effect of turmeric and black seed on pH value:
There is no significant effect of different treatment on pH value.
5. Effect of turmeric and black seed on volume of gastric
juice:
Significant decreases were observed in volume of gastric juice in
groups received (2.5% black seed, 5% black seed, 5% turmeric &
black seed) compared with control (+ve) group.
6. Effect of turmeric and black seed on total acidity:
All treated groups showed significant decreases in total acidity
compared with control (+ve) group7. Effect of turmeric and black seed on ulcer index:
All treated groups showed significant decreases in ulcer index
compared with control (+ve) group.
8. Effect of turmeric and black seed on feed intake,
weight gain and feed efficiency ratio:
Significant increases were observed in feed intake in group
received (5% turmeric, 2.5% black seed, 5% black seed, 5% turmeric
& black seed) compared with control they group.
Also, significant increases in feed efficiency ratio in group
received (5% turmeric, 2.5% black seed, 5% black seed, 2.5% turmeric
& black seed and 5% turmeric & black seed) compared with control
(+ve) group.
There were significant increases in body weights in all treated
group compared with control (+ve) group.
9. Effect of turmeric and black seed on organs weight:
Results showed significant increases in liver weights in group
received (2.5% turmeric & black seed, 5% turmeric & black seed)
compared with control (+ve) group. Also significant increases in
kidney weights in group received (5% turmeric & black seed)
compared with control (+ve) group.
10. Effect of turmeric and black seed on histopathology
examination:
Results showed improvement of histopathology examination of
stomach in all treated groups compared with control (+ve) group.
