Abstract

Floating famotidine-loaded calcium alginate beads were developed to prolong gastric residence time. Beads were prepared by different techniques using sodium alginate and pectin. Optimized beads were prepared using 2% sodium alginate solution, freeze drying technique and coated with Eudragit®RL100. Beads showed uniform size, high DEE%, remained afloat more than 24 hrs with no floating lag time. Beads exhibited sustained and prolonged drug release profiles while they are floating over the dissolution medium throughout the study for more than 12 hrs. The optimized formula, EUDR-F101 was prepared containing famotidine and QRT. The optimized formula showed excellent stability for up to 12 months and was subjected to in vivo study. The antiulcer activity of the optimized (Eudr-F101) formula was evaluated using rats. Indomethacin was used to induce gastric damage. The optimized formula was found to be more effective in treatment of gastric ulcers than the commercial famotidine tablets.
