Abstract

Bone marrow was obtained from long bones of Sprague Dawley rats. characterization of the isolated non adherent bone marrow stem cells was done by flow cytometric analysis .The isolated hematopoietic stem cells were differentiated into insulin-producing cells through 3 stage protocol. The formed clusters were stained crimson red by DTZ. The results of insulin release and C-peptide content indicated that, there was a stepwise increase in insulin release by formed clusters in response to increasing glucose concentrations. Mass balance calculations demonstrated that insulin released during glucose concentration was the result of endogenous synthesis. Flow cytometric analysis revealed that 16% of cells were positive for insulin. Gel electrophoresis and Q-PCR showed that clusters as well as rat pancreas expressed similar endocrine genes and transcription factors. Immunofluoresence analysis and immunocytochemical studies for the differentiated cells were positive for insulin and C-peptide.
