Abstract

Some of the most promising research focused in the field of medicine on the use of stem cells, these cells, by definition, is a non-differentiated cells and their intrinsic ability to regenerate addition, its high capacity to proliferate and produce a large number of new cells that have the ability to build new tissues.
Research is underway on the use of stem cells as ways to treat some diseases such as hematology. It has been prossible to obtain promising results in their use for the treatment of other diseases such as coronary heart disease, liver and brain. Characterized stem cells drived from bone marrow as a renewable source of many of the cells, such as some blood cells alo used as a source for many of the different tissues.
The bone marrow cells, as well as adipose tissue cells are of the most common sources to derive mesenchymal stem cells (MSC), while exporters are identical ability to differentiation (cartilage – bone – muscle – skeletal), but the cells drived from adipose tissue are easy accessible while there was no difference in morphology or immumohistochemical or the ability to isolate.
The process of converting mesenchymal stem cells into adipocyte is a complex process involving several steps through external stimulated is done by a mixture of stimulating hormones, which consists of a number of chemical compounds in certain proportions.
In this study we made a comparison between BMSCs and ATSCs isolated from male C57BL/6 mice, and then cultured in DMED-LG+10% FBS+1% antibiotic antimycotic. And expanded till passage 3 then the adipogenic differentiation media is added to the BMSCs and ATSCs to start differentiation.
Characterization of adipocyte derived from BMSCs and ATSCs was done by semi quantitative PCR for 5 genes ( peroxisome proliferator activated receptor- (PPAR- ), CCAAT/enhancer binding protein (C/EBP ), fatty acid binding protein 4 (FABP4),lipoprotein lipase (LPL) and beta actin ( -actin))
and staining the differentiated cells with oil red O stain which was specific to adipocyte and then measured the optical density of the extracted dye.
In conclusion, ADSCs are promising type of mesenchymal stem cells, and that according to its high proliferation rate, differentiation capability.but the problem in it as it may give some metabolism and debris which may affect on using stem cell as a treatment so it may be useful in the experimental study or in vitro that in vivo. In contrast, BMSCs give low yield and need more time to be used but its pure and can be used in vivo and in treatment that ADSCs. During the adipogenic process and before addition of differentiation media, the cells in the plate must be 70-80% confluent to give high RNA at the end of the process.
