Abstract Diabetes mellitus is a chronic disease that is associated with reduced life expectancy, significant morbidity and diminished quality of life. Diabetes was induced by injection of STZ in rats (50 mg/kg i.p.). Diabetic animals were treated with gliclazide (4.5 mg/kg p.o.), Morus alba extract (600 mg/kg p.o.) and Guar gum extract (200 mg/kg p.o.) alone or in combination for 14 days. The effect of these agents was studied on serum glucose, serum insulin and oral glucose tolerance test as well as glycosylated hemoglobin. Furthermore, oxidative stress biomarkers such serum malondialdehyde and blood glutathione levels as well as serum lactate dehydrogenase and blood superoxide dismutase activities were determined. In addition, serum urea level was measured. Finally insulin resistance, sensitivity and β-cell function were calculated. The following results were obtained: Effect of streptozotocin on the measured parameters: - Streptozotocin increased serum glucose level as well as increased HBA1c level and decreased serum insulin level. - Streptozotocin impaired oral glucose tolerance after oral glucose load, increased insulin resistance and decreased both insulin sensitivity and β-cell function. - Streptozotocin increased both serum MDA level and serum LDH activity and decreased blood SOD activity as well as GSH level. - Streptozotocin also raised serum urea level. Effect of gliclazide on the measured parameters: - Gliclazide decreased serum glucose level, increased serum insulin level and decreased HBA1c level. Summary and Conclusions __________________________________________________________________ 020 - Gliclazide corrected the impaired glucose tolerance as well as decreased insulin resistance and ameliorated both insulin sensitivity and β-cell function. - Gliclazide decreased both serum MDA level and LDH activity, also it elevated blood SOD activity as well as blood GSH level. - Administration of gliclazide decreased serum urea level. Effect of Morus alba extract on the measured parameters: - Morus alba extract decreased serum glucose, HBA1c levels and increased serum insulin level. - Morus alba extract corrected oral glucose tolerance as well as decreased insulin resistance and improved β-cell function. - Administration of Morus alba extract decreased MDA level, LDH activity and increased GSH level. - Morus alba extract did not affect SOD activity. - Morus alba extract reduced elevated serum urea caused by STZinduced diabetes. Effect of Guar gum extract on the measured parameters: - Guar gum extract decreased serum glucose level and increased serum insulin level, also Guar gum extract decreased elevation in HBA1c level. - Guar gum ameliorated oral glucose tolerance as well as decreased insulin resistance and improved β-cell function. - Guar gum extract reduced both serum MDA level and serum LDH activity. It also increased GSH level. - Guar gum extract did not affect SOD activity. - Guar gum extract decreased elevation in serum urea level caused by STZ-diabetic rats. Effect of combination of gliclazide and Morus alba extract on the measured parameters - This combination decreased serum glucose and increased serum insulin, it also decreased HBA1c level. Summary and Conclusions __________________________________________________________________ 022 - Combination of gliclazide and Morus alba extract corrected the impaired glucose tolerance test as well as ameliorated insulin sensitivity, β-cell function and decreased insulin resistance. - Concurrent administration of gliclazide and Morus alba extract nearly normalized MDA level and LDH activity. This combination significantly elevated blood SOD activity and GSH level. - This combination decreased serum urea level. Effect of combination of gliclazide and Guar gum extract on STZdiabetic rats: - This combination decreased serum glucose and HBA1c levels, coupled with an increase in serum insulin level. - Administration of gliclazide and Guar gum extract corrected impaired oral glucose tolerance. This combination also decreased insulin resistance and improved insulin sensitivity and β-cell function. - Combined administration of gliclazide and Guar gum extract decreased MDA level and nearly normalized the elevation in LDH activity caused by STZ-induced diabetic rats. This combination increased both SOD activity and GSH level. - This combination decreased serum urea level. Effect of Combination of Morus alba extract and Guar gum extract on STZ-diabetic rats: - This combination decreased serum glucose level and increased serum insulin level, also decreased HBA1c level. - Concurrent administration of Morus alba extract and Guar gum extract ameliorated the impaired glucose tolerance, decreased insulin resistance and increased insulin sensitivity as well as β-cell function. - The same combination decreased MDA level and LDH activity paralled to an increase in blood SOD activity and GSH level. - Administration of Morus alba extract and Guar gum extract lowered the elevation in serum urea level caused by STZ-induced diabetes. Summary and Conclusions __________________________________________________________________ 021 Combination of Morus alba extract and Guar gum extract produced an action greater than that of gliclazide alone on blood glucose level. Effect of combination of gliclazide, Morus alba extract and Guar gum extract in half doses on STZ-diabetic rats: - This combination nearly normalized serum glucose level, serum insulin level as well as HBA1c level. - Concurrent administration of gliclazide, Morus alba extract and Guar gum extract in half doses corrected the impaired glucose tolerance. It also decreased insulin resistance and improved both insulin resistance and β-cell function. - The same combination normalized oxidative stress biomarkers such as MDA level, LDH activity and SOD activity as well as increased GSH level. - Combined administration of gliclazide, Morus alba extract and Guar gum extract in half doses decreased serum urea level. Combination of Morus alba extract and Guar gum extract in half the doses with gliclazide in half dose synergized the action of gliclazide on blood glucose level. |
