Abstract

Colon cancer represents a major public health problem, accounting for more than one million new cases diagnosed each year and approximately a half million deaths worldwide. The aim of this study was to investigate the effect of radis Raphanus sativus) seeds polysaccharides in vitro by determining the anti-tumor activity of extracted polysaccharides against the different carcinoma cell lines. Also chemo-preventive, anti-tumor and antioxidant activity of extracted radish polysaccharides were studied in rats injected with dimethylhydrazine (DMH). Rats were subjected to subcutaneous injection by DMH with dose of 50mg/kg body weight for 8 consecutive weeks. Radish seeds polysaccharides (100 mg/kg body weight) were administrated for8 consecutive weeks before DMH injection and continued until the end of the experiment. Also polysaccharides were administrated for 8 consecutive weeks after DMH injection. The results obtained from the study revealed that chemical analysis of radish polysaccharides done by NMR and paper chromatography was b-galactan and the type of bond that exists between sugar residues is 1, 3 and 1, 6 glucosidic linkages. In vitro study, the potential cytotoxicity of radish seeds β- galactan had a notable cytotoxic activity on the different carcinoma cell lines with high cytotoxic activity on colon carcinoma cell line with. Radish b galactan was able to significantly decrease activities of serum ALT, AST and ALP. Serum cholesterol and triacylglycerols levels were lower in DMH-treated animals with radish b-galactan than that rats injected with DMH only. Moreover, serum glutathione reductase and glutathione- S- transferase activities were significantly increase in DMH-treated animals with radish seeds b-galactan than that rats injected with DMH only. DMH injected animals showed lymphoid follicle hyperplasia with infiltration the covering mucosa and dysplastic mucosa of colon. Also liver showed slightly disturbed lobular hepatic architecture with sinusoidal dilatation, mild focal steatosis and focal mild degenerative changes in the peripheral and middle zones, also showing mild focal nuclear dysplastic changes
