Abstract

We dealt in the thesis with the new synthetic organic heterocyclic imidazoles in laboratory of (prof. Dr. / A. A. Ali, Chemistry Department, Faculty of Science, Minia University), which we found out that they have a potent effect on prolonging Caenorhabditis elegans (C elegans) lifespan and they are: PHNID, BDHNID, CPHNID, MPHNID, PBHNID.
C elegans is a free living nematode worm and was deliberately selected as biological model organism because the lifespan of this worm is about 30 days, and also its simple nervous system which is one of the most important reasons to choose as a biological model organism. Celegans possessed particularly favorable characteristics for laboratory study and also has mechanisms that symmetry the biological processes in all the experimental animals species.
In our present study, C elegans cultivates under normal conditions in the laboratory, also in presence of the new imidazoles different concentrations comparing to the potent antioxidant proanthocyanidin, it it is iound that the concentrations 1 mM of PHNID, 1 ~M of BDHNID, 50 mM of PBHNID, 500 nM of MPHNID and 500 nM of CPHNID, has an obvious effect on prolonginging the wild-type N2 nematodes (Celegans) lifespan compared to the control group and this effect is similar to the effect of 76 flg/ml of the known antioxidant proanthocyanidine.
C elegans lifespan is affected with many factors. Oxidative stress resistance, it performs an important role in C elegans lifespan longevity. Consequently, the optimum mentioned concentrations from: PHNID, BDHNID, CPHNID, MPHNID, PBHNID, prolongs C elegans lifespan under standard laboratory conditions at the same time of reducing the oxidative stress.
In the actual reality, if these compounds have an effect as antioxidants, so the worm treatment with this compounds, reduce the production of ROS types and the advanced glycation end products.
So, we collected age· synchronized worms at day 5 and 20 , then we measured MDA and AGEs. If it is found from the results that worms treatment with the best chosen concentrations which are previously mentioned, reduce the amounts of MDA and AGEs.
Also, we measured the worm lifespan under high temperatures stress 30°C, 35°C, 40°C, it was found that the lifespan also prolongs worm treatment of chosen concentrations of imidazole compounds.
