Abstract

Urinary tract infection is the second most common type of infection in the body caused mainly by E.coli which is responsible for 75-90% f uncomplicated cystitis in younger women.(Foster et al.,2008).
Acquired antimicrobial resistance is a growing worldwide problem due to the increasing use of antimicrobials in human ,animals and agriculture. Sensitivity testing is used to select effective antibiotic which is used in treatment and control of infection especially when caused by pathogens that are often drug resistant (Schito et al.,2000).
The aim of this study was isolation and identification of E.coli and enterococcal isolates from urine samples , to determine the antimicrobial susceptibility for all E.coli and enterococcal isolates using three different methods of antibiotic sensitivity (disc diffusion, tube dilution and E-test)and to recommend the best method.
This study was conducted on 80 patients , Their age ranged from 20 to 60 years. They were 42 females (52.5 %) and 38 males (47.5%). 62 cases (77.5%) were outpatients while 18 cases(22.5%)were admitted in urology department in Benha University Hospital .
Antimicrobial susceptibility test by disc diffusion method was done for all E.coli isolates to different antimicrobials (ampicillin , cefotaxime , ciprofloxacin , chloramphenicol , gentamycin , cephalexin) and revealed that that the most sensitive antibiotic is gentamycin (93.75%) followed by ciprofloxacin(56.25%) while the least sensitive antibiotic is ampicillin(25%).
Antimicrobial susceptibility test by disc diffusion method was also done for all enterococcal isolates to different antimicrobials (ampicillin, cefotaxime,ciprofloxacin, chloramphenicol, gentamycin, cephalexin) and revealed that the most sensitive antibiotic is gentamycin (80%) followed by ciprofloxacin(70%) while the least sensitive antibiotic is cephalexin(20%).
Disc diffusion ,tube dilution and E-test were used to determine sensitivity of E.coli and enterococcal isolates to cefotaxime and ciprofloxacin.
E-test is better than disc diffusion as testing method of sensitivity of E.coli to cefotaxime and ciprofloxacin. This is done in comparison to tube dilution method which is taken as reference method.
