Abstract Duck plague considered the most dangerous virus disease affecting duck farms causing sever losses so the present work planned to prepare an effective inactivated DP vaccine by using different adjuvants and comparing between then to chose the most suitable one giving high and prolonged term immune response. To fulfill this goal the study include the followin -Successful propagation of DPV on Vero cell line, the highest titre 1010 TCID50/ml was obtained after 6 passages. -Toxicity test was done on new Montanide ISA 206 and Montanide ISA 563 oil adjuvants on Swiss male mice and resulted in non toxic oils. -Inactivation of seed virus by BEI, complete inactivation occurred after 15 hrs. Three batches of inactivated duck plague vaccine were prepared by using of Montanide ISA 206, Montanide ISA 563 and Aluminum hydroxide gel. Sterility test and safety test was done on the prepared vaccines revealed that prepared vaccines were sterile and safe for duck inoculation. Prepared vaccines were inoculated in duck groups, some receive one dose and other groups receive 2 doses by am apart. -Evaluation of the prepared vaccines and comparing them with each other was done by using serological tests SNA, ELISA. The prepared vaccines with Montanide ISA 206 oil adjuvart were high in antibody titre than prepared vaccine with Montanide ISA 563 and Aluminum hydroxide gel. The highest titre obtain d after 6 weeks of vaccination and remain high till reached up o 5ms post vaccination. At the end of the experiment at 8mpv a tibody titre still high and protective. -The ducks receive 2 doses of prepared inactivated DP accine exhibit higher titre of antibodies at 2 week post boosteri g and remain high till end of experiment 7ms boostering. -Montanide ISA 206 inactivated duck plague vaccine induce higher antibody titre than do by Montanide ISA 563 DP vaccine and Aluminum hydroxide gel vaccine. -Challenge test was done to all vaccinated groups giving 100% protection. |
