Abstract

In this study different immunization schedules for the production of anti-BCV IgY were tested and the produced yolk immunoglobulins were subjected to different purification protocols. The best immunization schedule found was the injection of hens with 2 doses 6 weeks apart then a booster after 10 weeks from the first dose, followed by monthly boostering. The immunoglobulins of collected yolk were purified either physically by direct freezing and thawing or chemically by the use of (HPMCP) alone or in combination with the precipitation of the protein using amm. sulphate. The obtained products were tested by ELISA,NT,TP,ZSTT,SDS-PAGE for purity and AGPT for precipitation character. Then the purified anti-BCV IgY ,used for the diagnosis of BCV infection in calf fecal samples, by Immuno-Dot-Blotting that gave the same sensitivity and specificity as the commercially available kit.
