Abstract

For evaluation the immune response of RB51 strain , Sera collected from cattle vaccinated with different Brucella vaccines commonly used in Egyptian field including RB51 are evaluated , tested and compared by Rose Bengal , IELISA using whole cell antigen and Western blot IELISA and Rose Bengal tests reavealed that the RB51 strain showed higher immune response than the other vaccinal strains while Western blot results reavealed that Rev - 1 has a great antigenic correlation and high immune reaction with field stain responsible for brucellosis in Egypt Whole cell proteins and OMP using SDS - PAGE profiles are used to differentiate between different brucella strains Whole cell proteins and OMP profiles were found to be comparable between different Brucella strains but to less extend PCR assays were done to characterize and differentiate the different Brucella strains included in this study The PCR assay using primer designated based on the sequencing of the IS711 can identify and differentiate Brucella as B abortus (RB51 , S19 and S2308) , B melitensis (Rev - 1 , 16M and three field strain which are B melitensis biovar 3 and B suis 1330 Also PCR assay using primer designated based on the sequencing of the IS711 - like element which disturb the wboA gene can identify and differentiate RB51 from its parent strain 2308 and also from any other Brucella strains
