Abstract

This study was aimed to identify the anesthetic effects of propofol as intravenous anesthetic solution in donkeys. This study was directed to study the effect of propofol alone or with other anesthetic and analgesic agents on heart rate, respiratory rate and the temperature, and also its effects on blood parameters blood pictures, liver and kidney function testes. The blood samples were collected before injection of each regimen and at, 15, 30, 60 minutes and at 24 hours.The present study was carried out on 20 donkeys.Fifteen donkeys of them (of both sexes), collected from the suburban of kalyobia governorates were used as an experimental model.The other five donkeys were chosen from the clinical cases that presented to the clinic of the faculty of veterinary medicine at Moshtohor to be castrated.The experimental model of animals were classified to five groups, 3 donkeys for each as follow Group I (Propofol / Propofol), the animals were injected with the initial dose of 2 mg/kg body weight of Propofol.The injection was very slowly over about 2 minutes. The anesthesia was maintained with propofol infusion rate of 0.2 mg/kg/minute diluted in 5% dextrose in a ratio of 1:4 respectively.Group II (Propofol / Xylazine hydrochloride), the animals were injected intravenously with xylazine HCL 2% in dose of 1mg/kg body weight.Once the animals become sedated, the initial dose of Propofol 2mg / kg body weight was injected and then it maintained by intravenous infusion of propofol in a dose of 0.2mg / kg body weight/ minute diluted in 5 % dextrose in ratio of 1:4 respectively.Group III (Propofol / chloral hydrate), the animals were injected slowly with 10% chloral hydrate solution in a dose of 3mg/ 50 kg body weight then the anesthesia was maintained by intravenous infusion of 0.2mg / kg/minute propofol diluted in 5 % dextrose in a ratio of 1:4 respectively.Group IV (Propofol / ketamine), the animals were injected intravenously with ketamine 10% in dose of 2.2 mg/kg. Once the animals became in a recumbent state, an initial dose of Propofol 2mg / kg body weight was also injected (The injection of each was very slowly).The anesthesia was maintained by propofol infusion 0.2mg / kg/minute diluted in 5 % dextrose in ratio of 1:4 respectively.Group V (Propofol / thiopental sodium), for induction of anesthesia, the animals were injected with a mixture of Propofol and thiopental.The dose of the Propofol 2mg/kg body weight was prepared and mixed with 2.5% thiopental solution in dose of 10 mg/kg body weight.Once the animals became sleep the anesthesia was maintained by Propofol infusion 0.2mg / kg/minute diluted in 5 % dextrose in ratio of 1:4 respectively The results showed that the propofol characterized by rapid onset of action and short duration of anesthesia with respiratory depressant action and increase in the heart rate in the group.I While using xylazine before propofol, it characterized by sedative action and the induction of the anesthesia was more rapid and the animals showed no excitation, with decrease in the respiratory rate.The using of chloral hydrate with propofol made the animals more calm and decrease the cost of used propofol.The respiratory rate showed decrease.The using of ketamine with propofol gave good anesthesia with normal non disturbed body function (haemodynamic stability The using of thiopental with propofol gives rapid onset, with decreased respiratory rate.The blood parameters were showed decrease in the RBCs then returned back after 24 hours and blood albumin showed decrease.In all groups, the recovery was rapid except the group III (chloral hydrate with propofol), and smooth with no complications.Conclusion Finally, the propofol provide smooth and rapid onset of action with short duration of anesthesia and rapid recovery. But it has high price and depressant effect on respiratory system However, we can solve its adverse effect via combination of propofol with other drugs to overcome its depressant effect on respiratory rate and its high price.We recommended the use of propofol with ketamine as in group IV, it give good anaesthesia with haemodynamic stability.
