Abstract

Ten isolates belonging to genus Rhizobium were isolated from healthy clover nodules, collected from different geographic areas of six governorates in Egypt, Sharkia, Suez, Qalubiya, Menofia, Esmailia and North Sinai. These rhizobial isolates were characterized at morphological, physiological, biochemical and molecular level. We used protein pattern and plasmid profile. from our results of protein pattern we found that all rhizobial isolates showed twenty seven protein bands ranged from (10 to 80 KD) which were not necessarily present in all isolates. By plasmid profile we found that all isolates contain large plasmid( sym plasmid) which may carry the genes responsible for nodulation . Also some isolates contain another small pasmid such as isolated No. 1,4,5 and 10 but there were isolates contain three plasmids such as isolate no. 3, 6,7 and 9.
Genetic relationships among rhizobial isolates were assessed by comparison between intrinsic antibiotic resistance profiles with protein profiles and we found that, except for one isolate, clusters based on antibiotic resistance profiles were similar to clusters obtained with protein profiles.
