Abstract

This investigation was carried out at the Experimental Farm of Sakha Agricultural Research Station and the Biotechnology Laboratory-Rice Research and Training Center(RRTC)- Agricultural Research Center (ARC), Kafrelsheihk Governorate, During the period from 2005/2006 to 2007/2008, with the cooperation of departments of Agronomy and Genetics, Faculty of Agriculture, Minoufiya University. The main objectives of this study were: 1- Studying the nature of inheritance of leaf rust disease resistance caused by Puccinia triticina and some of agronomic traits including grain yield and its components. 2- Detecting the number of resistant genes for leaf rust in the studied cultivars under field conditions. 3- Detecting the gene Lr37 of leaf rust resistance using Sequence Characterized Amplified Regions technique (SCAR) in the studied wheat cultivars. In 2005/2006 season, eight bread wheat parents were used in this investigation, four commercial cultivars namely, Giza 168, Sakha93, Sakha94 and Gemmeiza9 and four leaf rust monogenic lines,.i.e., Lr10, Lr19, L37 and Lr39 as testers, were sown. Top crosses were produced by using line * tester mating design to produce the 16 F1s hybrids. In 2006/2007 season, the parents and the 16 btained F1 s were sown to produce the F2 seeds and regenerating F12 hybrid seeds.In 2007/2008 season, the evaluation experiment was sown on late sowing date at 12 December to have good chance to be subjected to the pathogen inoculum (P. Triticina ) at the proper time. The 16 F1s, F2s and their eight parents arranged in a randomized complete block design with three replications. The spreader of experiment was inoculated with mixture of the prevalent leaf rust pathotypes.
