Abstract
This investigation presents studies on isolation of a pure line from duodenal oocysts, identification and justification of purity of this local isolate from Sharqia Government and evaluation of some common anticoccidial drugs applied in Egyptian farms on some of isolated Eimeria strains.
Single cell isolation and propagation was performed in one day old chicks and an E. maxima was identified using different parasitological parameters and was called Z1- E. maxima strain. Identification also includes shape that oval and largest oocyst and dimensions of width 21.00-35.00 µm, length 24.85 -43.05 µm and shape index 1.30 of oocysts, histological sections and sites of different schizonts and gametes in Lieberkuhn glands or in villi of the infected jejuni at different time post infection (30 hr,36 hr,42 hr,48 hr,60 hr,66 hr,96 hr,78 hr,72 hr108 hr,114 hr,120 hr,130 hr,144 hr,168 hr,192 hr,240 hr and 216 hr). No appearance (detection) of any developmental stage in caecal and / or in rectal tissues justified the purity of the local strain that isolated from duodenum after procedure of single oocyst isolation technique, in addition to all developmental (endogenous) stages of this isolate without exception were detected where the schizonts were located above host cell nuclei and all gametes were detected in the lamina propria (below host cell nuclei).
