Abstract

The present study is an endeavor for profound exploration of the influence of different antioxidants upon reproductive performance of adult male rats exposed to oxidative stress induced by cadmium chloride ” CdCl2 ”. Attention was also directed to estimate the most effective treatment that antagonizes testicular oxidative stress.
For this purpose, 180 adult male rats were equally divided into 9 groups; the 1st was considered as negative control administered distilled water. The 2nd ( positive control ) was treated with 1 / 100 LD50 of CdCl2. Rats of the remaining groups were administered 1 / 100 LD50 of CdCl2 followed 3 days later by 1 / 50 LD50 of either ascorbic acid, taurine and beta-mercaptoethanol or a combination of ascorbic acid and taurine, ascorbic acid and beta-mercaptoethanol, taurine and beta-mercaptoethanol as well as ascorbic acid, taurine and beta-mercaptoethanol, respectively for 2 successive months. At the end of the 8th week, individual blood samples were collected with their corresponding pituitaries. Sera and pituitaries, immediately after collection, were preserved in deep freeze at –20oC till the immunological assay of the studied hormones ( gametogenic hormone, interstitial cell stimulating hormone “ ICSH ” and testosterone “ T ” ). Moreover, individual tissue samples of the testis were obtained and prepared for histopathological studies as well as observation of spermatogenic stages. Meantime, corresponding individual semen samples were collected for semen evaluation. The obtained serum samples were also used to detect levels of total antioxidant capacity and Malondialdehyde ” MDA ” to evaluate the oxidant / antioxidant balance.
The obtained results revealed that :
Application of 1/100 LD50 CdCl2 for 8 successive weeks decreased pituitary and serum levels of gonadotropins and serum level of T, altered semen quality, decreased serum level of total antioxidant capacity, increased serum level of MDA and induced degeneration and necrosis of the lining spermatogenic cells.
On the other side, application of different antioxidants in rats under CdCl2 oxidative stress increased pituitary and serum levels of gonadotropins and serum level of T as well as improved semen quality, increased serum level of total antioxidant capacity, decreased serum level of MDA as well as restored testicular architecture to normal state.
In addition, the present study showed that the best improvement in male reproductive performance was achieved after supplementation with AA and taurine separately or in combination while the least improvement was obtained when β-ME was applied alone.
