Abstract

Mesenchymal stem cells are rare elements in the bone marrow stroma (2 to 5 cells per million nucleated cells), these cells play a critical role in the bone marrow stroma formation and regeneration, MSc are nowadays intensively studied not only for their haematopoeitic support but also for their ability to differentiate into specialized tissue as bone ,cartilage and others.
The aim of the present work was to estimate the percentage of CD44 positive stem cells,CD34 and CD45 in normal bone marrow.
In the present study, bone marrow aspirate was obtained from 20 patients attending the Main Alexandria University Hospital at department of Clinical Pathology between May 2008 to December 2008.
Patients were classified according to their hematological disorder into 3 groups:
Group I: Patients with hypersplenism
Group II: ITP patients.
Group III: Patients with NHL.
Bone marrow aspirate obtained from 20 patients was diluted, and then mesenchymal stem cells were isolated by adhesive technique and the level (%) of CD44, CD34 and CD45 were assessed by flow cytometery.
The assessment of CD44 marker gave positive results ( 20) with the 3 groups of patients [(group I = patients with hypersplenism) (group II = patients with ITP) (group III = patients with NHL)]
The assessment of CD34 and CD45 marker gave negative results (< 20%) with the 3 groups of patients [(group I = patients with hypersplenism) (group II = patients with ITP) (group III = patients with NHL)].
