Abstract

The present study was carried out on semen collected from healthy fertile bucks using rabbit artificial vagina to show the influence of different concentrations of calcium and calcium channel blocker (verapamil) on morphological changes in rabbit sperm which occurred concomitant to acrosome reaction. Also, the effect of calcium, verapamil, caffeine and prostaglandin F2a Ofl enzyrnac (aspartate aminotransferase, alanine aminotransferase) release from sperm cells was studied.
Rabbit spermatozoa were incubated in capacitating media (Brackett’s media) which contain different concentrations of Ca2+ (zero— 1.69mM —3.38 mM— 5.07 mM— 6.76 mM) for 3 hours at 3C. Aliquants from each concentration were taken at hourly intervals for checking the motility of spermatozoa, smears were stained by silver nitrate staining then examined microscopically by oil emersion lens to observe the morphological changes which occurred in the acrosome of rabbit sperm which include: intact acrosome, swollen acrosome, vesiculation of acrosome, peeling off of acrosome and appearance of equatorial segment.
A negative correlation between the concentration of C2 and the motility was observed. The highest motility percentage was observed in Ca2+ free media then sperm motility was decreased gradually with increasing the concentration of Ca2+.
