Abstract

Cytochrome P-450 enzymes are a key component of the mixed function oxidase The cytochrome P-450 are responsible for the metabolic activation of 3us endogenous and exogenous chemicals. The capacity of P-450 to metabolize array of substrates is due in part to the multiplicity of P-450 forms, many of possess overlapping substrate specificities. The process by which P-450 tes the metabolism of any givin substrate is by the insertion of a single oxygen fwhich is obtained from molecular oxygen, into the substrate. : In recent years, the role of reactive free radicals species, especially oxygen free 5, as a causative agents of cancer and aging has drawn much interest. Although re numerous defense mechanisms within the cell that have the capacity to limit [the leveles of reactive oxygen species and/ or the damage they induced, these oxygen species can escape and produce damage to DNA, protien and rated fatty acids.
| The mutagenic and carcinogenic potential of the environmental chemicals through Dacity to both directly bind to DNA molecules as well as induced single strand reaks due to the formation of oxygen free radicals has been hypothesized. The : P-450 to produce oxygen free radicals such as superoxide anion and hydrogen i are well documented. There is evidence suggesting the involvement of oxygen ficals in chemically mediated carcinogenesis. As most chemical procarcinogens vaied by cytochrome P-450, it was of interest to determine
