Abstract

It was found that 25 ul of phenoloxidase and 5.81 x 10 3 mole adrenaline (the substrate) are,suitable to be used in the reaction mixture. The optimum pH and temperature of the enzyme was 6.8 and 30°C, respectively. The enzyme has proved to reach its maximum activity after 10 min. incubation period.
On the other hand, it was found that 25 ul of AChE and 2.02x10-3 mole of ATCh (the substrate) are suitable to be used in the reaction mixture. The optimum pH and temperature of the enzyme was 8.0 and 25 °C, respectively. The enzyme has proved to reach its maximum activity after 10 min. incubation period.
Phenoloxidase and AChE activities of Apis mellifera were determined in both adult (head, thorax and abdomen) and larval stages (first, second, third, fourth and fifth instar). Head had more activity than thorax followed by abdomen. In the case of larval stages, fifth instar > fourth > third> second > first instar, respectively. This order was in phenoloxidase and AChE.
Dimilin, Methomyl, and their mixture (1:1) were tested in vitro on phenoloxidase adult (head, thorax and abdomen) and larval stages. Various concentration (10-9- 10-3 M) of the tested insecticides were applied to the reaction mixtures. The inhibitory effect of the tested insecticides were increased according to the concentration.
