Abstract

n This study, detection of CTV was proceeded by different techniques such as ELISA, IC/RT-PCR, Southern and dot blot hybridization. However, they showed some differences in their sensitivity; the IC/RT-PCR assay was more sensitive for virus detection than the immunological-based test. On the other hand, ELISA using PAbs was a very useful tool as a routine work for detection of CTV isolates, but ELISA using monoclonal antibodies was not specific enough for detection of all CTV isolates but, helpful for recognizing most isolates, which have related epitopes. In addition, we developed a cRNA probe specific to CTV-CP-ORF. The developed Dig-cRNA probe successfully detected CTV in citrus trees containing either the severe or mild strains or mixture of them with the same specificity and sensitivity. Dot blot hybridization could also be used as a routine work.
